| 背景信息 | TNF receptor superfamily member 8 (CD30) is a cell membrane protein and a member of the tumor necrosis factor receptor superfamily. It is primarily expressed on activated T and B cells but remains absent in resting lymphocytes. Upon binding its ligand, TNFSF8/CD30L, CD30 interacts with adapter proteins such as TRAF2 and TRAF5 to activate the NF-kappaB signaling pathway. This signaling cascade modulates diverse cellular processes, including the promotion of apoptosis, regulation of CD8 T cell proliferation, and protection against autoimmunity. CD30 is a well-established clinical biomarker and therapeutic target. It is highly expressed on Reed-Sternberg cells in Hodgkin lymphoma and neoplastic cells in anaplastic large cell lymphoma (ALCL), as well as other CD30-positive T-cell and B-cell lymphomas. Its clinical significance is further highlighted by its use in distinguishing embryonal carcinoma from seminoma and its role as the target for the antibody-drug conjugate brentuximab vedotin. |
