| 背景信息 | Caspase-1 (CASP1) is a cytosolic cysteine-aspartate protease and the central effector of the inflammasome complex. Synthesized as an inactive zymogen, it is recruited to inflammasome adaptors via its CARD domain, where it undergoes autoproteolytic activation. Active Caspase-1 cleaves the pro-forms of the inflammatory cytokines IL-1β and IL-18 into their biologically active secreted states, initiating potent inflammatory responses. Additionally, it cleaves gasdermin D to induce pyroptosis, a lytic form of cell death. Dysregulated Caspase-1 activity is a hallmark of autoinflammatory diseases, septic shock, and neurodegenerative conditions, driving chronic inflammation and tissue damage. |
