| 背景信息 | Serpin family A member 4 (SERPINA4), also known as kallistatin, is a secreted glycoprotein and a member of the serpin superfamily that serves as a potent inhibitor of tissue kallikrein. By forming a stable complex with kallikrein, it blocks the generation of kinins, thereby regulating vasodilation and vascular homeostasis. Beyond its protease inhibitory activity, SERPINA4 modulates inflammation, angiogenesis, and oxidative stress by signaling through receptors such as LRP6, nucleolin, and KLF4. It exhibits pleiotropic effects, including the suppression of VEGF-induced endothelial proliferation and the inhibition of TNF-alpha-mediated NF-kappaB activation. Clinically, reduced levels of SERPINA4 are associated with the progression of colorectal cancer, where it functions as a tumor suppressor and prognostic biomarker by inhibiting cell proliferation and invasion. It is also implicated in the pathogenesis of diabetic retinopathy. |
