| 背景信息 | CD40 ligand (CD40LG), also known as CD154, is a type II transmembrane glycoprotein expressed primarily on activated T cells. It belongs to the tumor necrosis factor (TNF) superfamily and plays a critical role in immune regulation by binding to its receptor, CD40, on antigen-presenting cells such as B cells, dendritic cells, and macrophages. This interaction triggers signaling pathways that regulate immunoglobulin class switching, antibody production, and the activation of NF-κB and MAPK pathways. CD40LG is essential for T-cell-mediated "help" in B-cell activation and differentiation, enabling the production of IgG, IgA, and IgE antibodies. It also contributes to cytokine production and enhances immune responses through integrin-mediated signaling. Mutations in the CD40LG gene are associated with X-linked hyper-IgM syndrome, a rare immunodeficiency disorder characterized by impaired antibody class switching and increased susceptibility to infections. Abnormal CD40LG expression has also been linked to autoimmune diseases and certain cancers. |
