| 背景信息 | Myristoylated alanine rich protein kinase C substrate (MARCKS) is a ubiquitous membrane-associated protein that regulates actin cytoskeleton plasticity, cell motility, and vesicular trafficking. It features a basic effector domain that binds PIP2 at lipid rafts and crosslinks F-actin, serving as a reversible molecular switch. Upon phosphorylation by PKC or binding to Ca2+/calmodulin, MARCKS translocates from the plasma membrane to the cytosol, facilitating processes such as chemotaxis, adhesion, and exocytosis in various cell types, including immune cells. MARCKS interacts with multiple signaling pathways, including PI3K-AKT and TLR, to drive inflammatory responses and cytokine release. Clinically, MARCKS is implicated in the pathogenesis of lung diseases, endothelial dysfunction in vascular disorders, and neuronal dendritic defects in neurodegeneration. It also plays a significant role in promoting cancer cell proliferation and fibrosis. |
