| 背景信息 | Major histocompatibility complex, class I, F (HLA-F) is a non-classical MHC class I molecule that forms a heterodimer with beta-2-microglobulin. Unlike classical MHC molecules, HLA-F primarily localizes to the endoplasmic reticulum and Golgi, with limited surface expression. It features a unique open-ended peptide-binding groove that allows for the presentation of unconventional long or phosphorylated peptides. HLA-F exists in both peptide-bound forms and as empty open conformers, which differentially interact with immune receptors. Peptide-bound HLA-F engages the inhibitory receptor LILRB1 to promote immune tolerance, particularly at the maternal-fetal interface. Conversely, open conformers can bind activating receptors like KIR3DS1 on natural killer cells to trigger cytokine release. These interactions allow HLA-F to modulate NK and T cell responses in various antiviral, antitumor, and inflammatory contexts. |
