| 背景信息 | Ribonucleotide reductase regulatory subunit M2 (RRM2) is the small subunit of the ribonucleotide reductase complex, an essential enzyme that catalyzes the conversion of ribonucleotides to deoxyribonucleotides. This process provides the necessary precursors for DNA synthesis, repair, and replication. RRM2 features a dinuclear non-heme iron center required for free radical generation and its subsequent oxidoreductase activity. Its expression is strictly regulated by the cell cycle, peaking during the S phase to support DNA replication. Post-translational modifications, such as acetylation and phosphorylation, further modulate its stability and enzymatic activity. Clinically, RRM2 is frequently overexpressed in various cancers, including choriocarcinoma, where it drives malignant behaviors and promotes epithelial-mesenchymal transition via the JAK2/STAT3 pathway. Additionally, mutations in the RRM2 gene have been linked to Diamond-Blackfan anemia 20. |
