| 背景信息 | BMI1 proto-oncogene, polycomb ring finger (BMI1) is a key catalytic component of the Polycomb repressive complex 1 (PRC1), which maintains the transcriptional repression of various genes through H2AK119 ubiquitination. It features an N-terminal RING finger domain essential for E3 ubiquitin ligase activity and a central helix-turn-helix domain for DNA interaction. BMI1 is a critical regulator of stem cell self-renewal, cell cycle progression, and senescence, primarily by repressing the INK4a/ARF locus. This repression bypasses senescence barriers and modulates the p53 and pRb pathways. Additionally, BMI1 participates in DNA double-strand break repair and interacts with Wnt and Notch signaling. Clinically, BMI1 is overexpressed in numerous malignancies, including leukemia and breast cancer, where it drives oncogenesis, promotes epithelial-mesenchymal transition, and serves as a prognostic biomarker for therapy resistance and tumor progression. |
