| 背景信息 | Thrombospondin 2 (THBS2) is a disulfide-linked homotrimeric matricellular glycoprotein primarily secreted by fibroblasts, endothelial cells, and macrophages. It functions as a critical modulator of cell-to-cell and cell-to-matrix interactions within the connective tissue. THBS2 inhibits angiogenesis by binding to the CD36 receptor and regulates pericellular protease activity through its interaction with matrix metalloproteinase 2 (MMP2). It also influences the bioavailability of growth factors and the migration of mesenchymal cells, thereby maintaining matrix integrity. Clinically, mutations in the THBS2 gene are associated with classic-like Ehlers-Danlos syndrome type 3, characterized by joint hypermobility and skin hyperextensibility, as well as intervertebral disc disease. Conversely, THBS2 has been shown to play a protective role in rheumatoid arthritis by suppressing synovial inflammation and preventing the formation of ectopic lymphoid structures. |
