| 背景信息 | Keratin 9 (Cytokeratin-9) is a type I intermediate filament protein primarily expressed in the suprabasal layers of the palmoplantar epidermis. It features a central alpha-helical rod domain flanked by glycine- and serine-rich head and tail domains. KRT9 functions by forming heterodimers with type II keratins, which assemble into 10-12 nm filaments essential for maintaining the mechanical strength and cytoskeletal integrity of stress-bearing skin. It also plays a role in the terminal differentiation of keratinocytes. Clinically, mutations in the KRT9 gene are the primary cause of epidermolytic palmoplantar keratoderma (EPPK). This dominant-negative disorder is characterized by severe thickening and blistering of the skin on the palms and soles, resulting from the fragility of the keratin filaments and subsequent collapse of the epidermal structure under mechanical stress. |
