| 背景信息 | Msh homeobox 2 (MSX2) is a transcription factor belonging to the muscle segment homeobox family, characterized by a homeodomain that enables sequence-specific DNA binding. It functions primarily as a transcriptional repressor, playing a crucial role in craniofacial morphogenesis by balancing cell survival and apoptosis in neural crest-derived cells. MSX2 is a key regulator of bone development, where it represses ALPL promoter activity and antagonizes the effects of DLX5 during osteoblast differentiation. It also suppresses osteocalcin and helps prevent the mineralization of ligaments and tendons by inhibiting Runx2. The protein interacts with components of the general transcription factor TFIIF through its core suppressor domain. Expressed in the bone marrow, brain, and muscle, MSX2 is essential for proper limb patterning and the regulation of cell growth through the RAS signaling pathway. |
