| 背景信息 | Tyrosine kinase with immunoglobulin like and EGF like domains 1 (TIE1) is an endothelial-specific cell surface receptor essential for the development and maintenance of the vascular and lymphatic systems. It functions as a context-dependent modulator of TEK/Tie2 signaling through the formation of Tie1/Tie2 heterodimers or Tie1 homodimers. Although considered an orphan receptor, TIE1 regulates the activation of Tie2 by angiopoietins, with its own ectodomain cleavage by VEGF serving to relieve Tie2 inhibition. This mechanism is critical for maintaining microvessel integrity and endothelial stability during angiogenesis. Clinically, TIE1 is implicated in the pathogenesis of severe edema, lymphatic defects, and tumor-associated angiogenesis, and it plays a recognized role in the progression of atherosclerosis. |
