| 背景信息 | Protein phosphatase, Mg2+/Mn2+ dependent 1A (PPM1A) is a ubiquitously expressed magnesium-dependent serine/threonine phosphatase localized to both the cytoplasm and the nucleus. It functions as a critical negative regulator of multiple signaling pathways by catalyzing the removal of phosphate groups from diverse substrates, thereby opposing kinase activity. Key targets include YAP/TAZ in the Hippo pathway, SMAD2/3 in TGF-beta signaling, Axin in Wnt signaling, and various MAP kinases, which collectively control cell proliferation, differentiation, and cell cycle progression. PPM1A also activates the p53 tumor suppressor to induce G2/M arrest and apoptosis. Dysregulation of this enzyme is linked to various pathologies; for instance, decreased expression promotes renal fibrosis through the impaired inhibition of SMAD3 signaling, while altered activity affects wound healing, inflammation, and neovascularization. |
