| 背景信息 | Keratin 2 (Cytokeratin-2e) is a type II basic keratin protein that plays a fundamental role in maintaining the cytoskeletal integrity of epidermal cells. It features a central alpha-helical rod domain that facilitates the formation of coiled-coil heterodimers with type I keratins. KRT2 is specifically expressed in the suprabasal layers of the stratum spinosum within stratified epithelia. Clinically, mutations in the KRT2 gene are associated with several skin disorders characterized by disrupted filament assembly. These include ichthyosis bullosa of Siemens, which presents with superficial blistering and epidermal fragility, and the Curth-Macklin type of ichthyosis hystrix, which causes severe palmoplantar keratoderma and hyperkeratosis. These conditions underscore the protein's essential function in providing mechanical resistance to the upper layers of the epidermis. |
