| 背景信息 | Transforming growth factor beta receptor 1 (TGFBR1), or ALK5, is the type I receptor serine/threonine kinase of the TGF-beta signaling pathway. Upon ligand binding, it forms a complex with type II receptors and phosphorylates SMAD proteins to regulate cell proliferation, differentiation, and apoptosis. TGFBR1 exerts tumor-suppressive effects by inhibiting uncontrolled cell growth, though dysregulated signaling can promote tumorigenesis in advanced cancers. Germline loss-of-function mutations in TGFBR1 cause Loeys-Dietz syndrome, a connective tissue disorder characterized by arterial aneurysms and craniofacial anomalies. Somatic mutations are also found in various cancers. |
