| 背景信息 | Hepatocyte nuclear factor 4 alpha (HNF4A) is a highly conserved nuclear receptor that functions as a homodimeric transcriptional regulator of hepatic gene expression. It plays a vital role in the endodermal-to-hepatocyte transition and the maintenance of metabolic homeostasis, including cholesterol, fatty acid, and glucose metabolism. Structurally, HNF4A features a DNA-binding domain with two zinc-finger modules that recognize specific direct repeat DR1 elements and a ligand-binding domain that binds endogenous fatty acids to maintain structural stability. Its activity is further modulated by post-translational modifications, such as phosphorylation at Ser78, which influences DNA binding and nuclear localization. |
