| 背景信息 | TNF superfamily member 9 (4-1BBL) is a type II transmembrane protein expressed on antigen-presenting cells, including B cells, macrophages, and dendritic cells. It typically assembles into a homotrimeric, bell-shaped configuration characteristic of the TNF superfamily, featuring a jellyroll beta-sandwich architecture. Upon binding to its cognate receptor, 4-1BB, the ligand undergoes conformational changes that facilitate immune activation and T cell costimulation. This interaction is predominantly mediated by the cysteine-rich domains of the receptor. Unlike its murine counterpart, which forms stable homodimers, human 4-1BBL functions as a trimer, highlighting important species-specific variations in the assembly and signaling of TNF superfamily ligands. |
