| 背景信息 | Green fluorescent protein (GFP) tag is a 238-amino-acid barrel-shaped protein derived from the jellyfish Aequorea victoria, widely utilized as a fusion tag for real-time protein visualization in living cells. Its intrinsic fluorescence arises from an autocatalytically formed chromophore (Ser65-Tyr66-Gly67) shielded within an 11-stranded beta-barrel structure. GFP absorbs blue/UV light and emits bright green fluorescence, requiring proper folding and oxygen for activity. Engineered variants, such as enhanced GFP (EGFP), offer improved brightness and folding efficiency for mammalian expression. When fused to the N- or C-termini of target proteins via flexible linkers, GFP allows for the detailed study of protein localization, trafficking, and dynamics with minimal perturbation to native function. |
