| 背景信息 | Glutamate ionotropic receptor AMPA type subunit 1 (Glutamate Receptor 1) is an integral membrane protein that functions as a ligand-gated cation channel, mediating fast excitatory synaptic transmission in the central nervous system. It assembles into tetrameric complexes featuring an extracellular amino-terminal domain for dimerization and a clamshell-like ligand-binding domain that responds to L-glutamate. This binding induces conformational changes in the transmembrane domain to open the ion pore. The receptor is essential for neural communication, memory, and synaptic plasticity. Mutations in the encoding gene are associated with various neurodevelopmental disorders, including intellectual disability, autism spectrum disorder, and schizophrenia, as well as neurological conditions such as epilepsy and Alzheimer’s disease. |
