| 背景信息 | Rho GTPase activating protein 25 (ARHGAP25) is a Rac-specific GTPase-activating protein predominantly expressed in hematopoietic cells. It negatively regulates Rho-type GTPases, particularly Rac1, by accelerating GTP hydrolysis to an inactive GDP-bound state. This activity modulates actin cytoskeletal dynamics, cell polarity, and migration. ARHGAP25 contains pleckstrin homology and GAP domains and is involved in critical hematopoietic processes, including phagocytosis, leukocyte transendothelial migration, and B-cell development. In clinical contexts, ARHGAP25 often acts as a tumor suppressor; its downregulation is associated with metastasis in colorectal and pancreatic cancers. Conversely, its upregulation in certain conditions, such as alveolar rhabdomyosarcoma, has been linked to increased cellular invasion. |
