| 背景信息 | Phosphatidylinositol-4,5-bisphosphate 3-kinase catalytic subunit alpha (PI3 Kinase p110 alpha) is the 110 kDa catalytic component of class IA PI3K heterodimers. It pairs with regulatory subunits to phosphorylate PIP2 into PIP3, thereby triggering downstream signaling pathways essential for cell growth, survival, and metabolism. The protein's modular structure includes a Ras-binding domain and a kinase domain, which are activated upon the release of inhibitory regulatory domains. PI3K p110 alpha is a critical mediator of insulin signaling and hepatic metabolism. Gain-of-function mutations in the PIK3CA gene are highly prevalent in human cancers, driving approximately 30% of solid tumors, including breast and colorectal cancers. Additionally, these mutations are associated with the PIK3CA-related overgrowth spectrum (PROS), which includes conditions such as CLOVES syndrome. |
