| 背景信息 | TXK tyrosine kinase (TXK) is a non-receptor tyrosine kinase belonging to the Tec family that plays a vital role in regulating adaptive immune responses. It is primarily involved in the development, differentiation, and activation of T cells and NKT cells, often functioning redundantly with ITK. Upon T-cell receptor stimulation, TXK translocates to the plasma membrane where it undergoes autophosphorylation to achieve full activation. Once active, it phosphorylates PLCG1, triggering intracellular calcium release and the nuclear translocation of NFAT. TXK also acts as a Th1-specific transcription factor, forming a complex with PARP1 and EEF1A1 to bind the IFNG promoter and drive the production of interferon-gamma. Through these mechanisms, TXK modulates the actin cytoskeleton and cytokine production, serving as a key coordinator of type II interferon-mediated pathways and Th1 cell function. |
