| 背景信息 | Cathepsin O (CTSO), encoded by the CTSO gene, is a lysosomal cysteine protease belonging to the peptidase C1 family. It functions in intracellular protein degradation, antigen processing, and extracellular matrix remodeling. CTSO is synthesized as an inactive zymogen (proenzyme) that undergoes proteolytic cleavage in acidic lysosomal environments to become active. The mature enzyme has a molecular weight of approximately 29.5 kDa and exhibits optimal activity at acidic pH. It is expressed ubiquitously, with higher levels observed in osteoclasts, suggesting a role in bone resorption. Structural features include a signal peptide, propeptide region, and a catalytic domain with conserved cysteine residues essential for enzymatic activity. Dysregulation of CTSO is linked to pathological conditions. Overexpression has been implicated in tumor progression, facilitating cancer cell invasion and metastasis through extracellular matrix degradation. Reduced CTSO activity may contribute to bone disorders such as osteoporosis due to impaired osteoclast-mediated bone turnover. Additionally, altered expression is associated with inflammatory and autoimmune diseases. |
