| 背景信息 | NK2 homeobox 1 (Thyroid Transcription Factor 1) is a homeodomain-containing transcription factor essential for the development and physiological function of the thyroid, lungs, and brain. It activates the transcription of critical thyroid-specific genes, including thyroglobulin and thyroperoxidase, and regulates the expression of surfactant proteins in the lung that are necessary for respiratory function. In the central nervous system, NKX2-1 is vital for the development and migration of interneurons. Clinically, mutations in the NKX2-1 gene result in brain-lung-thyroid syndrome, characterized by congenital hypothyroidism, infant respiratory distress, and movement disorders. In oncology, NKX2-1 is a widely used diagnostic marker; its loss is associated with the progression of thyroid cancer and the development of mucinous lung adenocarcinoma, where the reactivation of embryonic gastric programs leads to poor clinical outcomes. |
