| 背景信息 | Troponin T3, fast skeletal type (TNNT3) is the tropomyosin-binding subunit of the troponin complex specifically expressed in fast-twitch skeletal muscle. It plays a central role in muscle contraction by regulating calcium-mediated interactions between thick and thin filaments within the sarcomere. TNNT3 undergoes developmental regulation through alternative splicing, transitioning between isoforms to tune muscle contractility and calcium sensitivity during maturation. Clinically, mutations in TNNT3 are associated with distal arthrogryposis multiplex congenita, including Sheldon-Hall syndrome and distal arthrogryposis types 2B1 and 2B2. These mutations disrupt the ability of the troponin complex to properly regulate filament binding, leading to the joint contractures and skeletal abnormalities characteristic of these disorders. |
