| 背景信息 | NOP2 nucleolar protein (NOP2) is a highly conserved S-adenosyl-L-methionine-dependent RNA methyltransferase that catalyzes the C5-methylation of cytosine 4447 in 28S rRNA. This modification is essential for efficient pre-rRNA processing and the biogenesis of the 60S ribosomal subunit. NOP2 also regulates ribosome assembly through non-catalytic interactions with box C/D snoRNAs, promoting the recruitment of U3 and U8 snoRNAs to pre-90S particles. Beyond its role in ribosome biogenesis, NOP2 is involved in the positive regulation of cell proliferation and p53-mediated signaling. Clinically, NOP2 is overexpressed in various malignancies, including high-grade serous ovarian cancer, where it acts as an oncogenic factor. It promotes tumor cell proliferation, migration, and invasion through m5C-dependent regulation of the cAMP pathway, and its high expression levels are significantly correlated with poor patient prognosis. |
