| 背景信息 | Notch receptor 1 (NOTCH1) is a single-pass transmembrane protein that plays a crucial role in cell fate determination, differentiation, proliferation, and survival. It is part of the evolutionarily conserved Notch signaling pathway, which regulates interactions between physically adjacent cells. NOTCH1 contains an extracellular domain with multiple EGF-like repeats and an intracellular domain that acts as a transcription factor when cleaved. The protein functions as a receptor for membrane-bound ligands such as Jagged-1, Jagged-2, and Delta-1, activating gene expression upon ligand binding. NOTCH1 is involved in various developmental processes, including T-cell differentiation, angiogenesis, and neurogenesis. Mutations in the NOTCH1 gene are associated with several diseases, including aortic valve disease, Adams-Oliver syndrome, and various types of cancer, particularly T-cell acute lymphoblastic leukemia and chronic lymphocytic leukemia. |
