| 背景信息 | IQ motif containing GTPase activating protein 1 (IQGAP1) is a large cytoplasmic scaffold protein that integrates multiple signaling pathways to regulate actin cytoskeleton organization, cell adhesion, and polarity. Despite its homology to Ras GTPase-activating proteins, IQGAP1 does not stimulate GTP hydrolysis; instead, it binds and stabilizes the active, GTP-bound forms of Cdc42 and Rac1. Through its diverse domains, it interacts with a wide array of partners, including F-actin, E-cadherin, β-catenin, and components of the MAPK signaling cascade such as B-Raf and ERK1/2. These interactions allow IQGAP1 to coordinate cytoskeletal remodeling with cell-cycle progression and migration. Pathologically, IQGAP1 is frequently overexpressed in various cancers, where it drives oncogenic signaling, invasion, and metastasis, making it a significant target for therapeutic intervention. |
