| 背景信息 | Inhibitor of DNA binding 4 (ID4) is a member of the ID family of proteins characterized by a conserved helix-loop-helix (HLH) domain but lacking a basic DNA-binding region. It functions as a dominant-negative regulator by dimerizing with basic HLH (bHLH) transcription factors, thereby preventing them from binding to DNA and activating target genes. ID4 is the longest member of its family and possesses unique N-terminal and C-terminal domains that enable flexible protein-protein interactions. During development, ID4 is essential for maintaining oligodendrocyte progenitor cells and preventing premature differentiation. In oncogenesis, high expression of ID4 is linked to the maintenance of stem-cell-like states and increased plasticity in melanoma and glioma. It is specifically associated with chemo-resistance in glioblastoma through the modulation of SOX2 and ABC transporters. |
