| 背景信息 | Neuronal pentraxin 2 (NPTX2), also known as NARP, is a synaptic protein encoded by the NPTX2 gene and is part of the neuronal pentraxin family, which is structurally related to C-reactive protein. NPTX2 functions as an immediate-early gene product that plays a pivotal role in excitatory synapse formation and synaptic plasticity by clustering AMPA-type glutamate receptors at established synapses. It is secreted in response to neuronal activity and can aggregate excitatory receptors, facilitating long-term neuronal plasticity and potentially modifying cellular properties underlying these processes. NPTX2 forms complexes with other pentraxins, such as NPTX1 and neuronal pentraxin receptor (NPR), and is involved in calcium-dependent binding and receptor internalization, which may protect against excitotoxicity. Clinically, NPTX2 is highly upregulated in Parkinson’s disease (PD) tissues, particularly in the substantia nigra, and is a novel component of Lewy bodies and Lewy neurites, suggesting a role in PD pathology and dopaminergic neuron death. |
