| 背景信息 | Leptin receptor (LEPR) is a transmembrane protein encoded by the LEPR gene, belonging to the cytokine receptor superfamily, and is widely expressed in various tissues, with particularly high levels in the hypothalamus. Structurally, LEPR consists of an extracellular domain for leptin binding, a transmembrane domain, and a cytoplasmic domain essential for intracellular signal transduction, notably through activation of JAK tyrosine kinase and STAT transcription factors. The receptor’s extracellular region contains a WSXWS motif, characteristic of cytokine receptors, which is important for proper folding and function. Upon leptin binding, LEPR initiates signaling cascades that regulate energy homeostasis, appetite, and metabolism, primarily by promoting satiety and reducing hunger. Mutations or deficiencies in LEPR disrupt these pathways, leading to persistent hunger, overeating, and severe obesity, as well as associated endocrine disorders such as hypogonadism and infertility. The receptor’s function is also modulated by related proteins, such as LEPROT, which negatively regulate its cell surface expression and leptin responsiveness. |
