背景信息Keratin 6A (Cytokeratin-6A) is a type II intermediate filament protein that provides mechanical resilience and structural integrity to stratified epithelia, including the epidermis of the palms and soles, oral mucosa, and hair follicles. It typically forms heteropolymers with type I keratins, such as KRT16 or KRT17. Beyond its structural role, Keratin 6A is involved in wound healing by promoting keratinocyte migration and contributes to innate immunity through the release of C-terminal antimicrobial peptides. It is dynamically expressed in response to epithelial stress and serves as a marker in certain aggressive cancers. Mutations in the KRT6A gene are the primary cause of pachyonychia congenita type 1, a rare autosomal dominant disorder characterized by hypertrophic nail dystrophy, painful palmoplantar keratoderma, and oral leukokeratosis.,Keratin 6C (Cytokeratin-6A) is a type II intermediate filament protein essential for the structural integrity and mechanical resilience of epithelial cells in the skin, hair, and nails. It forms dense cytoskeletal networks by assembling into heteropolymeric filaments with type I keratins. Highly expressed in stratified epithelia, Keratin 6C shares significant sequence identity with its paralogs KRT6A and KRT6B. Clinically, mutations in the KRT6C gene, such as p.Asn172del, cause focal palmoplantar keratoderma and are associated with subtypes of pachyonychia congenita. Additionally, the overexpression of Keratin 6C has been implicated in cancer progression, where it promotes epithelial-mesenchymal transition, migration, and immune evasion in colorectal and lung adenocarcinomas, often correlating with a poor clinical prognosis.
