| 背景信息 | Tryptophan 2,3-dioxygenase (TDO2) is a heme-dependent cytosolic enzyme that catalyzes the first and rate-limiting step of the kynurenine pathway. It facilitates the oxidative cleavage of L-tryptophan into N-formylkynurenine, thereby regulating systemic tryptophan levels. Structurally, TDO2 forms a homotetramer with a conserved active site architecture that exhibits cooperative substrate binding and stereoselectivity. The enzyme plays a pivotal role in metabolic homeostasis and immune regulation. In oncology, elevated TDO2 expression is a well-recognized mechanism of tumor immune evasion; by depleting local tryptophan and producing kynurenine metabolites, it suppresses T-cell responses and promotes a tolerogenic environment. This activity correlates with poor prognosis in patients with melanoma and esophageal carcinoma. Furthermore, TDO2 is implicated in the activation of fibroblast-like synoviocytes in autoimmune arthritis, contributing to chronic inflammation and tissue destruction. |
