| 背景信息 | Cartilage oligomeric matrix protein (COMP) is a secreted, pentameric extracellular matrix glycoprotein of the thrombospondin family. It is highly expressed in articular cartilage, tendons, and ligaments, where it binds to collagens I, II, and IX to facilitate collagen fibrillogenesis and maintain matrix organization. COMP also supports chondrocyte attachment and mechanotransduction. Heterozygous mutations in the COMP gene cause the autosomal dominant chondrodysplasias pseudoachondroplasia (PSACH) and multiple epiphyseal dysplasia (MED), characterized by the intracellular retention of mutant protein and impaired bone growth. Additionally, wild-type COMP serves as a robust clinical biomarker in serum and synovial fluid for cartilage destruction in osteoarthritis and rheumatoid arthritis, reflecting disease progression and joint damage. |
