| 背景信息 | NUMB endocytic adaptor protein (NUMB) is a critical regulator of intracellular trafficking, asymmetric cell division, and cell signaling attenuation. It functions as an adaptor protein that facilitates both clathrin-dependent and -independent endocytosis, as well as early endosome fusion and receptor recycling. NUMB interacts with a variety of partners, including alpha-adaptin, Eps15, and EHD family proteins, to promote the vesicular docking and sorting of cargoes such as transferrin and LDL. A primary mechanism of NUMB is the antagonism of Notch signaling through the Itch-mediated ubiquitination and endocytosis of Notch1. Additionally, it modulates TrkB signaling for BDNF-induced chemotaxis and regulates APP trafficking. Through these diverse interactions, NUMB maintains cellular homeostasis and directs differentiation. Dysregulation of NUMB is associated with neuronal progenitor defects and has been implicated in the pathogenesis of neurodegenerative conditions such as Alzheimer's disease. |
