| 背景信息 | Vascular endothelial growth factor C (VEGFC) is a secreted signaling glycoprotein that serves as a primary regulator of lymphangiogenesis and angiogenesis. It is synthesized as a precursor that requires stepwise proteolytic processing by CCBE1 and ADAMTS3 to release the mature, fully active homodimer. Structurally, VEGFC contains a central VEGF homology domain with a cystine-knot motif that mediates high-affinity binding to its receptors, VEGFR-2 and VEGFR-3. The activation of these receptors promotes the proliferation, migration, and survival of lymphatic endothelial cells, making VEGFC essential for the development of the lymphatic system during embryogenesis. Clinically, dysregulated VEGFC signaling is strongly associated with the development of lymphedema and impaired wound healing. In oncology, its overexpression is a major driver of aberrant lymphangiogenesis, which facilitates the lymphatic metastasis of various tumors and contributes to chronic inflammatory diseases. |
