| 背景信息 | DNA ligase 3 (LIG3) is an ATP-dependent enzyme that catalyzes the joining of DNA strand breaks, playing unique roles in both the nucleus and mitochondria. In the nucleus, LIG3 often complexes with XRCC1 to participate in base excision repair and alternative non-homologous end-joining pathways. A distinct mitochondrial isoform serves as the sole DNA ligase in the organelle, making it indispensable for mitochondrial DNA maintenance and cell survival under oxidative stress. LIG3 utilizes a zinc-finger domain and a catalytic core to bridge and ligate DNA ends. Biallelic mutations in LIG3 cause mitochondrial DNA depletion syndrome 20, a severe condition characterized by neurogastrointestinal and myopathic phenotypes. Its expression levels are also studied as biomarkers in various malignancies, reflecting its central importance in maintaining genomic and mitochondrial integrity. |
