| 背景信息 | Histamine N-methyltransferase (HNMT) is a cytoplasmic enzyme that plays a crucial role in the metabolism of histamine, a biogenic amine involved in various physiological processes, including neurotransmission and immune responses. HNMT catalyzes the transfer of a methyl group from S-adenosyl-L-methionine to histamine, converting it into Nτ-methylhistamine, which is then further metabolized or excreted. This enzymatic activity is particularly important in the central nervous system (CNS), where HNMT is the primary enzyme responsible for terminating the neurotransmitter actions of histamine, thus regulating neural pathways related to arousal, appetite, and sleep-wake cycles. Dysregulation of HNMT has been linked to several diseases; mutations in the HNMT gene have been associated with nonsyndromic autosomal recessive intellectual disability. Additionally, altered HNMT activity may contribute to conditions such as allergies and neurodegenerative diseases. |
