| 背景信息 | PDZ and LIM domain 4 (PDLIM4), also known as RIL, is a cytoskeletal adaptor protein that plays a critical role in organizing the actin cytoskeleton and recruiting signaling molecules. It features a PDZ domain for actin association and a LIM domain for the recruitment of kinases and protein phosphatases. PDLIM4 facilitates actin binding, alpha-actinin binding, and protein homodimerization, localizing to structures such as actin stress fibers, Z-discs, and adherens junctions. Functionally, it influences actin dynamics, cell migration, and cell cycle control by promoting G1 phase arrest. In clinical contexts, PDLIM4 acts as a tumor suppressor in prostate cancer, where its expression is frequently reduced, leading to increased growth and clonogenicity. Additionally, human orthologs have been implicated in the pathogenesis of osteoporosis. |
