| 背景信息 | ST13 Hsp70 interacting protein (Hsc70 Interacting Protein HIP) is a 418-amino-acid adaptor protein that facilitates the association between HSP70 and HSP90 chaperones. It contains a TPR domain that interacts with the ATPase domain of HSC70, stabilizing it in the high-affinity ADP-bound state. This mechanism delays nucleotide exchange, thereby enhancing protein folding, preventing protein aggregation, and aiding in the assembly of the glucocorticoid receptor complex. ST13 is ubiquitously expressed and functions as a co-chaperone that attenuates the Hsp70 catalytic cycle. Clinically, ST13 is recognized as a tumor suppressor, with significant downregulation observed in colorectal carcinoma. Furthermore, mutations or dysregulation of this protein are linked to neurodegenerative conditions, specifically spinocerebellar ataxia autosomal recessive 16 and cerebellar ataxia type 48, highlighting its importance in maintaining proteostasis. |
