| 背景信息 | Nuclear receptor ROR-gamma (RoRgT) is a ligand-dependent nuclear receptor belonging to the NR1F subfamily that regulates transcription by binding to ROR response elements. It features a modular structure including a DNA-binding domain and a ligand-binding domain that accommodates oxysterols and other sterol derivatives. The RORgamma-t isoform is a master regulator of the immune system, driving the differentiation of Th17 cells and the production of pro-inflammatory cytokines like IL-17. The full-length RORgamma isoform also influences circadian rhythms and metabolic processes in the liver. Dysregulated ROR-gamma activity is strongly linked to the pathogenesis of autoimmune diseases, such as multiple sclerosis and psoriasis, making it a significant therapeutic target for inverse agonists designed to suppress immune-mediated inflammation. |
