| 背景信息 | Tetratricopeptide repeat domain 3 (TTC3) is an E3 ubiquitin-protein ligase encoded within the Down syndrome critical region on chromosome 21. It regulates key signaling pathways by catalyzing the formation of polyubiquitin chains, most notably targeting phosphorylated Akt for nuclear degradation to control cell survival and cycle progression. TTC3 also influences neuronal differentiation through interactions with citron kinase and modulates TGF-beta signaling by degrading SMURF2. Clinically, dysregulation of TTC3 is associated with the developmental defects observed in Down syndrome. Furthermore, specific variants of TTC3 have been linked to the pathogenesis of Alzheimer’s disease and other neurodevelopmental abnormalities, likely through its effects on actin cytoskeleton organization and PI3K-Akt signaling pathways. |
