| 背景信息 | PMS1 homolog 2, mismatch repair system component (PMS2) is an essential DNA mismatch repair protein that maintains genomic integrity by correcting errors during DNA replication. It functions as a key component of the MutLα complex, which initiates eukaryotic mismatch repair through its intrinsic endonuclease activity. The protein interacts with proliferating cell nuclear antigen (PCNA) through a specific hexapeptide motif to activate its endonuclease function, which is critical for tumor suppression. Beyond its role in mismatch repair, PMS2 contributes to the DNA damage response, genetic recombination, and the generation of antibody diversity through class switch recombination in B cells. Mutations in the PMS2 gene, such as the E705K variant, impair its endonuclease activity and are strongly associated with increased genomic mutation rates and cancer predisposition, including conditions such as Turcot syndrome. |
