| 背景信息 | GDNF family receptor alpha 1 (GFRA1) is a glycosylphosphatidylinositol (GPI)-anchored cell-surface co-receptor for glial cell line-derived neurotrophic factor (GDNF) and neurturin. It forms a signaling complex with the RET receptor tyrosine kinase to promote neuronal survival, differentiation, and morphogenesis within the central, peripheral, and enteric nervous systems. Structurally, GFRA1 is a cysteine-rich protein that assembles with GDNF in a 2:2 stoichiometry; it can also form higher-order decameric assemblies that mediate synaptic cell adhesion independently of RET. Clinically, GFRA1 is essential for proper organogenesis, and its dysfunction is implicated in congenital renal malformations, including renal hypodysplasia and bilateral renal agenesis. Additionally, due to its critical role in the development of the enteric nervous system, it is recognized as a candidate gene for Hirschsprung disease. |
