| 背景信息 | Ubiquitin associated and SH3 domain containing A (UBASH3A) is a cytoplasmic and nuclear adaptor protein predominantly expressed in T cells. It features a ubiquitin-associated (UBA) domain, an SH3 domain, and a phosphatase domain. UBASH3A functions as a negative regulator of T-cell receptor (TCR) signaling and endocytosis by binding ubiquitin, dynamin, and CBL. This interaction inhibits CBL-mediated downregulation of receptor tyrosine kinases, leading to the surface accumulation of activated receptors such as TCR and EGFR. By modulating T-cell activation thresholds, UBASH3A plays a significant role in immune homeostasis. Genetic variants affecting its expression or function are associated with susceptibility to autoimmune conditions, including type 1 diabetes and other immune-mediated disorders. |
