| 背景信息 | ARF GTPase 6 (ARF6) is a small GTP-binding protein that coordinates vesicular trafficking and actin cytoskeleton dynamics at the plasma membrane. It cycles between active GTP-bound and inactive GDP-bound states to regulate clathrin-independent endocytosis and the recycling of transmembrane proteins, including integrins and E-cadherin. These processes are essential for cell migration, adhesion, and the maintenance of epithelial and vascular barrier functions. ARF6 also facilitates membrane lipid remodeling and interacts with effectors like MMP14 to promote cellular invasion. Dysregulation of ARF6 signaling is linked to vascular instability and the progression of aggressive cancers, where its activation drives tumor growth, motility, and metastasis. Consequently, ARF6 serves as a critical regulator of both physiological membrane trafficking and pathological cancer cell behavior. |
