| 背景信息 | RAP1A is a protein encoded by the RAP1A gene, belonging to the Ras-related protein family. It shares approximately 50% amino acid identity with classical RAS proteins but has distinct structural features, notably a threonine at the 61st amino acid position instead of glutamine found in RAS proteins. RAP1A counteracts the mitogenic function of RAS by competitively interacting with RAS GAPs and RAF. It plays a crucial role in integrin activation, particularly in the context of thrombin-stimulated, integrin-dependent glioblastoma cell proliferation and tumor growth. RAP1A's interaction with integrins is mediated through effectors like RAPL and RIAM, which are involved in integrin clustering and adhesion. The protein's localization to membranes where integrins are found allows it to modulate integrin affinity and clustering directly. RAP1A's role in signaling pathways underscores its importance in cellular processes and its potential involvement in diseases such as glioblastoma. |
