| 背景信息 | FKBP prolyl isomerase 1A (FKBP12), or FKBP1A, is a cytosolic immunophilin with peptidyl-prolyl cis-trans isomerase activity that assists in protein folding. It is best known for binding the immunosuppressants FK506 and rapamycin; these drug-protein complexes inhibit calcineurin and mTOR, respectively. Through these interactions, FKBP12 plays a pivotal role in modulating T-cell activation, adaptive immunity, and cell growth pathways. Additionally, FKBP12 regulates intracellular calcium release channels, including ryanodine receptors, thereby influencing cardiac contractility and neuronal signaling. Perturbation of FKBP12 function is associated with mechanisms underlying cardiomyopathy and neurotoxicity. |
