| 背景信息 | Tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein zeta (YWHAZ) is a highly conserved adapter protein belonging to the 14-3-3 family. It regulates diverse signaling pathways by binding to phosphoserine and phosphothreonine motifs on partner proteins, influencing cell proliferation, migration, and neuronal maturation. YWHAZ modulates the activity of key factors such as RAC1, TFEB, and IRS1, the latter of which affects insulin sensitivity. In oncology, YWHAZ functions as an oncogene and is frequently upregulated in lung, breast, and prostate cancers, where it promotes epithelial-mesenchymal transition, metastasis, and chemoresistance through interactions with the ERK, AKT, and β-catenin pathways. Additionally, specific pathogenic variants of YWHAZ are associated with intellectual disability and cognitive defects, highlighting its importance in both developmental neurology and cancer progression. |
