| 背景信息 | Glypican 5 (GPC5) is a cell-surface heparan sulfate proteoglycan attached to the plasma membrane via a GPI anchor. It functions as a critical co-receptor that modulates major signaling pathways, most notably the Hedgehog (Hh) and fibroblast growth factor (FGF) pathways. Through its highly sulfated glycosaminoglycan chains, GPC5 binds both Sonic Hedgehog and its receptor Patched-1 at the primary cilia, thereby enhancing Hh-mediated cell proliferation and migration. In clinical contexts, GPC5 is frequently overexpressed in rhabdomyosarcoma, where it acts as a driver of tumor growth. Its role in developmental signaling and its association with various cancers underscore its importance in both normal morphogenesis and oncogenic transformation. |
